Vixen is based on the DC Character Mari McCabe / Vixen, a costumed superhero crime-fighter with the power to mimic the abilities of any animal that has ever lived on Earth. The series is set in the same fictional universe as Arrow, The Flash, and Legends of Tomorrow.

Priča nas vodi daleko u prošlost i dubine Afričkog kontinenta, gde ratnik Tantu od Anansi pauka dobija totem, koji onome ko ga nosi daruje sve moći životinjskog carstva, pod uslovom da te moći koristi kako bi štitio nevine. Uz pomoć tog totema, Tantu postaje prvi afrički heroj. Totem se prenosio sa genercije na generaciju, sve dok nije došao do porodice Mekejb.
Meri Mekejb saznaje za legendu o totemu od svoje majke. Njenu majku ubijaju lovokradice, pa Meri ostaje da živi sa ocem Ričardom, seoskim sveštenikom. Kasnije joj oca ubija general Maksai, njegov polu-brat, koji krade Tantu totem. Meri tada odlazi za Ameriku i dobija posao supermodela u jednoj njujorškoj agenciji. Stečeni novac koristi kako bi putovala svetom, pa se nekoliko godina kasnije vraća u Afriku i uzima totem od svog ujaka. Zahvaljujući moćima koje daruje totem, Meri postaje superheroina po imenu Vixen.
